The GCN4 gene encodes a positive regulator of unlinked amino acid by synthetic genes. The GCN4 gene is itself regulated by amino acid availability at the translational level. The GCN4 transcript has a 5' leader that is about 600 nucleotides long and contains four small open reading frames. An internal deletion in the leader, which eliminates the four small ORFs, abolishes regulation of the fusion enzyme and results in constitutively derepressed fusion enzyme levels which are about 40 fold higher than the repressed GCN4-plus-lacZ enzyme level.
